Effect of buffer system on the Xanthine oxidase activiting in the preparation procedure

Abstract
Xanthine Oxidase is terminal enzyme of purine body Catabolism and that product is uric acid. Thus uric acid is converted to allantoin by uricase. In this study, Xanthine Oxidase enzyme was prepared to various homogenizing buffer system and the enzyme activity is tested in each other buffer system. Activity of Xanthine Oxidase was not changed in 0.25 M Sucrose homoginizing buffer but 0.1M phosphate buffer, 0.1M Tris and 0.1M Tris(containing 1.15% kcl) buffer systems were significantly decreased. These results suggested that 0.25M Sucrose is the most available buffer among the various homogenizing buffer.
